Tirzepatide
In the clinical trial, patients lost on average up to 22.5% of their body weight depending on dose level. About one in three patients lost 25% or more of their body weight.
Tirzepatide works by mimicking the functions of certain hormones in the body involved in appetite regulation and glucose control. This results in reduced hunger, improved insulin sensitivity, and effective weight loss. Moreover, Tirzepatide helps stabilize blood sugar levels, making it a suitable treatment for patients with type 2 diabetes.
Tirzepatide is typically suitable for adults who have obesity or struggle with weight loss through diet and exercise alone. It's also an option for individuals with type 2 diabetes who need better blood sugar control.

Semiglutide
In the clinical trial, patients taking Semaglutide achieved an average 15% reduction in body weight after titrating up to the maximum target dose. One in three lost 20% or more of their body weight.
Semaglutide works by mimicking the function of a hormone in your body that regulates appetite and food intake, helping to reduce cravings and control portion sizes. In addition, it helps keep blood sugar levels stable, making it a suitable treatment for patients with type 2 diabetes.
Semaglutide treatment is suitable for adults who are overweight or obese and struggle to lose weight through diet and exercise alone. This therapy can also be used for individuals with type 2 diabetes to improve blood sugar control.

Contraindications
The use of medications for weight management is indicated for those patients who have a BMI of 30 or higher or a BMI of 27 or higher with other medical conditions such as high blood pressure, diabetes, or high cholesterol. Prescribing medications for patients not fitting these criteria, is considered “off label” and not “FDA approved.” Therefore, the potential risks vs. benefits may be great.
You should not take GLP-1 agonist medications if:
You have a personal or family history of “Thyroid C-Cell Tumors”. More specifically, a personal or family history of: Medullary thyroid carcinoma, MTC, (a specific, rare type of thyroid cancer) Multiple Endocrine Neoplasia syndrome type 2 (MEN-2, a rare, genetic disorder that can cause tumors in the thyroid gland and other glands) In light of the potential risk for MTC with use of GLP-1 medications (like semaglutide or tirzepatide), patients should be aware of symptoms of thyroid tumors (e.g. a mass in the neck, elevation of calcitonin, difficulty swallowing (dysphagia), difficulty breathing (dyspnea), persistent hoarseness, or other; and to seek appropriate evaluation for such signs or symptoms if present.
You are pregnant or plan to become pregnant while taking this medicine. GLP-1 medications may cause fetal harm. When pregnancy is recognized, discontinue GLP-1 medications. Discontinue GLP-1 medications at least 2 months before a planned pregnancy. You have a history of Pancreatitis or have a condition that may make Pancreatitis more likely to occur. Acute pancreatitis, including fatal and non-fatal hemorrhagic or necrotizing pancreatitis, has been observed in patients treated with GLP-1 receptor agonists (like semaglutide and tirzepatide).
You have a history of gallbladder disease. While not an an absolute “contra-indication” to GLP-1 medications, treatment with GLP-1 medications (i.e. semaglutide or tirzepatide) was associated with an increased occurrence of cholelithiasis (gallstones) and cholecystitis (gallbladder inflammation).
You have kidney disease or are on dialysis. There have been post-marketing reports of acute kidney injury and worsening of chronic kidney failure, which in some cases required hemodialysis, in patients treated with semaglutide.
